SETAC Presentation: OECD 301B Ready Biodegradation Test Design

Test substances can be labeled “Ready Biodegradable” if 60% of their organic carbon is converted to CO2 in the presence of activated sludge during a 10-day period of the 28-day OECD 301B Guideline. Should the test substance be deemed non-ready biodegradable, the inherent biodegradability is assessed using the OECD 302 series.

Observing mineralization of test substance can be difficult with the OECD 302 series tests. Smithers Research Director, Sean McLaughlin, presents results from a modified OECD 301B that uses increasing amounts of inoculum, yet is not swamped by endogenous CO2 evolution from the additional activated sludge. This CO2 Evolution Test can be designed with activated sludge inoculum concentrations up to 100 mg/L to evaluate difficult to test compounds for their potential inherent biodegradability.
