2023 ACS Presentation - in vitro comparative metabolism of chemicals that are nominally a gas

Volatile organic chemicals (VOCs) pose technical difficulties for in vitro comparative metabolism testing due to their low solubility and high volatility. Current in vitro methodologies are typically designed for highly soluble and low volatility organic molecules. The aim of the work described was to develop appropriate methodologies for VOCs using a [14C] labelled, small, highly volatile (b.p.12°C), active ingredient.

Applied at nominal concentrations of 10, 50 and 100 µM VOC incubated with liver S9 fractions at ca 37°C with secondary cofactors. The incubation times for all species and concentrations were 0, 60 and 180 minutes in triplicate. Samples were treated with ice-cold ethanol to terminate the metabolic processes.

The aqueous mixture was assayed by LSC and metabolic profile generated using HPLC-RAM. The concentration of VOC in the headspace was quantified by GC-FID. Average mass balance (%TAR) across all incubation times ranged between 88.4% to 85.1%. This presentation discusses the experimental design, the challenges of analysis, results and viability of comparative metabolism experiments with [14C]-VOCs.
